Retatrutide is a single molecule engaging three receptors: GIP, GLP-1 and glucagon. Its pharmacology was taken from discovery to clinical proof of concept in 2022[2]; in a phase 2 trial it produced substantial, dose-dependent weight reduction in adults with obesity[1], and it improved glycaemic control in a randomised phase 2 trial in type 2 diabetes[6].
GHK-Cu is the copper complex of glycyl-L-histidyl-L-lysine, a tripeptide naturally present in human plasma. It has been studied as a modulator of multiple cellular pathways in skin regeneration — collagen synthesis, remodelling and antioxidant defence[3] — and, in a rat model of ACL reconstruction, transiently improved healing outcome[4].
Melanotan II is a cyclic synthetic analogue of alpha-melanocyte-stimulating hormone that activates melanocortin receptors. It is used as a pharmacological scaffold in melanocortin-receptor research[5], and in rodent models melanocortin-receptor agonism with Melanotan II decreased appetitive responding for food[7].
Together, the three vials give researchers three independent pharmacological axes: incretin/glucagon receptor signalling (retatrutide), copper-peptide chemistry (GHK-Cu) and melanocortin receptor signalling (Melanotan II).
